Heterobifunctional protein crosslinkers contain two different functional groups, which have their own reactivity and selectivity. These crosslinkers can be used in a one step process or can be used to create activated proteins or antibodies, which can often be preserved and reacted with the second biomolecule in a separate step.

Heterobifunctional crosslinking reagents are often used for conjugate formation, where one protein or antibody is reacted with an amine reactive group (such as NHS or Sulfo-NHS), and the other protein is reacted with the other end of the crosslinker, which has a different selectivity (such as a maleimide group). These crosslinkers have been used in the formation of hapten carrier proteins and in preparing enzyme-antibody conjugates.
