|DTSSP is a cleavable protein crosslinking reagent that contains a disulfide moeity in its eight atom spacer arm. DTSSP is a membrane impermeable, homobifunctional crosslinker that is reactive towards amine (amino) functional groups. In the pH range of 7-9, this reaction is particularly selective for primary amine groups, resulting in the formation of stable amide linkages. Phosphate buffers and phosphate buffered saline are typically used for crosslinking reactions involving DTSSP, although others non-amine containing buffers may also be used.
Once the reaction between DTSSP and proteins or peptides is complete, the disulfide-containing crosslinked protein can be cleaved via reduction of the crosslinkers disulfide bond using a reducing agent, such as 2-mercaptoethanol (CovaChem 11301-10x1) or DTT (CovaChem 11302-10x5). Dithiobis(sulfosuccinimidyl propionate) (DTSSP) is the water soluble version of Lomant's Reagent (DSP). Both DSP and DTSSP have been used extensively in protein crosslinker applications as well as other scientific applications involving forming a stable linkages between the disulfide group and metal surfaces, while leaving the NHS functional groups available to react with other nucleophiles. This application can be useful for immobilizing proteins or other molecules to the surface of gold beads or other foil.
Due to the lipophobic nature of the sulfonate groups on DTSSP, this crosslinker does not permeate the cell membrane. This feature makes DTSSP particularly useful for crosslinking membrane proteins on the surface of the cell without interfering with intercellular proteins.
