Once the reaction between DTSSP and proteins or peptides is complete, the disulfide-containing crosslinked protein can be cleaved via reduction of the crosslinkers disulfide bond using a reducing agent, such as 2-mercaptoethanol (CovaChem 11301-10x1) or DTT (CovaChem 11302-10x5). CovaChem's technical support team is readily available to assist with any individual optimization.
Dithiobis(sulfosuccinimidyl propionate) (DTSSP) is the water soluble version of Lomant's Reagent (DSP). Both DSP and DTSSP have been used extensively in protein crosslinker applications as well as other scientific applications involving forming a stable linkages between the disulfide group and metal surfaces, while leaving the NHS functional groups available to react with other nucleophiles. This application can be useful for immobilizing proteins or other molecules to the surface of gold beads or other foil.
Due to the lipophobic nature of the sulfonate groups on DTSSP, this crosslinker does not permeate the cell membrane. This feature makes DTSSP particularly useful for crosslinking membrane proteins on the surface of the cell without interfering with intercellular proteins.
